  1. Production, purification and characterization of mouse monoclonal antibodies

    • Service 1

      Conventional immunization protocol on Balb/c mice using a costumer-supplied antigen (immunogen). We usually use 3 female mice per immunization, but it is possible to increase the number of animals if deemed necessary. The minimum amount of antigen for the completion of phase 1 and phase 2 of this procedure is 1 mg. Intraperitoneal antigen injection is followed by spleen extirpation and fusion of mouse spleen cells with myeloma cells. Antigen-reactive clones are normally selected from about 100 clones using an ELISA screening. Unless otherwise requested, the three most promising clones are selected for further analysis and subclonig.

      Unless specifically requested, the selected clones from the first screening are subcloned once using standardized protocols, i.e. further subcloning can be performed upon request.

      If desired, it is possible to perform the first and the second screening (clone selection) on customer-provided tissues or cells, using other methods than ELISA, such as immunohistochemistry (included tissue microarrays) or immunocytochemistry (including cell-ELISA).

      Antibody labelling service is offered based upon conventional/commercial labelling kits, which can either arbitrarily selected by COMT (fluorescent labeling) or specifically requested by the customer. We offer tagging of immunoglobulins purified in-house or by the customer (in this case a prior check of the purity and integrity of the molecules is needed) with enzymes, fluorescent dyes, biotin, digoxigenin or other conventional tags.

      Evaluation the specificity and the functional properties of monoclonal antibodies, produced by us or customer-supplied, using ELISA, Western blotting, immunoprecipitation, immunohistochemical and immunocytochemical assays, flow cytometry and in vitro cellular assays.

      It is also possible to request development of specific immuneassays such as “capture ELISA” or non-isotopic “sandwich ELISA” with antibodies, control molecules and calibration agents provided by the customer or by COMT.   2. Preparation of tissue microarrays and immunohistochemical screening services

    • Production of low- and high-density tissue microarrays (TMAs), starting from samples provided by the customer in the form of unprocessed samples (i.e. fresh or fixed samples) or paraffin-embedded samples, or by accessing the COMT internal archival material. Composition and content of TMAs generated in-house are subject to current availability of material.
